Reconstitution

Reconstitution is the process of dissolving a lyophilized peptide in a solvent such as bacteriostatic water to prepare it for laboratory use.

In practice the chosen solvent is added slowly down the inner wall of the vial and the solution is swirled gently rather than shaken, because mechanical agitation and foaming can promote peptide aggregation. The appropriate solvent depends on the compound’s solubility: bacteriostatic water is common for multi-withdrawal vials, while sterile or mildly acidified water may suit poorly soluble peptides.

Once dissolved, the resulting concentration is simply the mass of peptide divided by the volume of solvent added, and the solution is usually divided into aliquots to limit freeze-thaw cycling. Careful reconstitution technique preserves structural integrity for downstream in-vitro work.
