The GIP receptor (GIPR) is a class B G protein-coupled receptor for glucose-dependent insulinotropic polypeptide that signals through the Gs/cAMP pathway. It is the second canonical incretin receptor.
Like GLP-1R, GIPR couples to Gs and raises intracellular cAMP on activation, driving PKA and Epac signaling in cell models. It is a shared target of dual agonists such as tirzepatide and triple agonists such as retatrutide.
In vitro, tirzepatide behaves at GIPR much like the native GIP peptide, which is a defining feature of its imbalanced dual-agonist profile.
