The GLP-1 receptor (GLP-1R) is a class B G protein-coupled receptor that binds glucagon-like peptide-1 and signals through the Gs/cAMP pathway. It is a primary target of incretin-mimetic research peptides.
On agonist binding, GLP-1R couples to the stimulatory G protein Gs, activating adenylyl cyclase and raising cyclic AMP (cAMP), which in turn engages protein kinase A (PKA) and Epac. It can also recruit beta-arrestin, a pathway used to characterize biased agonism in vitro.
GLP-1R is one of the two canonical incretin receptors and a shared target of dual and triple agonists studied in receptor-pharmacology assays.
