A triple agonist is a single molecule engineered to activate three receptors at once. In incretin-class research this means simultaneous agonism at the GLP-1, GIP, and glucagon receptors.
Triple agonists such as retatrutide (GLP-3RT) build on dual agonists by adding glucagon-receptor (GCGR) activity to GLP-1R and GIPR engagement. All three are Gs-coupled class B GPCRs, so the shared cAMP output can be studied as an integrated signal in cell models.
