The glucagon receptor (GCGR) is a class B G protein-coupled receptor for the hormone glucagon that signals primarily through the Gs/cAMP pathway. It is the third receptor target added by triple agonists.
GCGR is a close structural relative of the incretin receptors and shares their Gs/adenylyl-cyclase/cAMP output; in some cell backgrounds it also couples to Gq-linked calcium signaling. It is studied most often in hepatocyte-derived models.
Adding GCGR activity to GLP-1R and GIPR engagement is what defines a triple agonist such as retatrutide (GLP-3RT) and is a focus of multi-receptor pharmacology research.
