Tirzepatide is a synthetic 39-amino-acid peptide studied in vitro as a dual agonist of the GLP-1 receptor (GLP-1R) and GIP receptor (GIPR). It is supplied for laboratory research use only; not for human or veterinary use.
Tirzepatide activates two Gs-coupled class B GPCRs with a single molecule, raising intracellular cAMP through adenylyl cyclase. In cell assays it shows an imbalanced potency profile, behaving much like native GIP at GIPR while acting as a weaker partial agonist at GLP-1R.
Structurally it carries Aib substitutions for DPP-4 resistance and a C20 fatty-diacid moiety that promotes albumin binding and extended exposure in preclinical models.
